ORDERED MECHANISM OF BINDING OF PHOSPHORYLASE KINASE AND GLYCOGEN PHOSPHORYLASE b TO GLYCOGEN

چکیده
The kinetics of the interaction of rabbit skeletal muscle phosphorylase kinase with glycogen has been studied using turbidimetric method (40 mM Hepes, pH 6.8, and 8.2; 20◦C). The binding of phosphorylase kinase with glycogen occurs only in the presence of Ca and Mg . According to the kinetic data, phosphorylase b favors the binding of phosphorylase kinase with glycogen. This conclusion is supported by the direct study of the influence of phosphorylase b on the binding of phosphorylase kinase with glycogen by sedimentation velocity in an analytical ultracentrifuge. A model for adsorbtion of phosphorylase b and phosphorylase kinase on the surface of the glycogen particle has been proposed. The model explains how phosphorylase b favors the tighter binding of phosphorylase kinase with glycogen.

Calcium-dependent phosphorylation of glycogen synthase by phosphorylase kinase.
Purified preparations of rabbit skeletal muscle glycogen synthase (specific activity 6 to 42 units/mg of protein) were found to be contaminated with calciumdependent kinase(s) which catalyzed the phosphorylation of glycogen phosphorylase b and glycogen synthase. In a series of glycogen synthase preparations there was a correlation between the phosphorylation rates of glycogen synthase and of ad...
متن کاملRole of the Sarcoplasmic Reticulum in Glycogen Metabolism
Sarcoplasmic vesicles and beta-glycogen particles 30-40 mmicro in diameter were isolated from perfused rabbit skeletal muscle by the differential precipitation-centrifugation method. This microsomal fraction was subjected to zonal centrifugation on buffered sucrose gradients, in a B XIV Anderson type rotor, for 15 hr at 45,000 rpm in order to separate the two cytoplasmic organelles. Zonal profi...
متن کاملActivation of hepatic glycogen phosphorylase b in vivo by sodium sulphate in normal (Wistar) and phosphorylase b kinase-deficient (gsd/gsd) rats.
Sulphate ions have been known for some years to enhance the activity of hepatic glycogen phosphorylase b in vitro. Here we report that intravenous injections of 4.92 mmol of Na2SO4/kg body wt. to rats induced marked hepatic glycogenolysis in vivo, accompanied by polyuria, glycosuria and a mild hyperglycaemia. These effects were observed both in normal (Wistar) rats and in gsd/gsd rats that lack...
متن کاملPhosphorylase kinase of the liver: deficiency in a girl with increased hepatic glycogen.
Studies of a child with glycogenosis revealed an increased concentration of glycogen and low phosphorylase activity in her liver. Using mixtures of homogenates of the patient's liver and of normal liver, we found the low phosphorylase activity to be caused by a deficiency of phosphorylase kinase and not of hepatic phosphorylase. The fact that phosphorylase activity was restored to normal values...
متن کاملGlycogen as a fuel for skeletal muscle.
The utilization of glycogen by skeletal muscle was, at one time, one of the central problems of biochemistry. Attention focused on the most dramatic property of the system: the very rapid glycogenolysis that occurs synchronously with muscle contraction. This phenomenon now has a generally accepted explanation : the increase in Ca2+ that induces muscle contraction also activates phosphorylase ki...
